Comparing Mass Photometry And CDMS For AAV analysis

A recent study by Eduard Ebberink and his colleagues at Utrecht University compared mass photometry and charge-detection mass spectrometry (CDMS) techniques for their ability to distinguish AAV loading distributions and evaluate adeno-associated virus (AAV) capsid content. The research is significant for AAV production and development. Both methods were used to assess AAVs from two different serotypes, packed or empty, and confirmed variability in AAV loading. The study found that different production methods resulted in differing packaging efficiencies. Both techniques were deemed excellent for measuring DNA packaging in AAVs, but mass photometry was highlighted as a simpler, faster method, capable of measuring AAV mass under physiological conditions. Researcher Eduard Ebberink intends to continue utilizing mass photometry in his work.
